I was wondering for their lore are we allowed to include ideas from the mythological definition of a Nephilim and other influences as well?

@CosmicFalcon yeah, go for it! The subspecies started because I thought it was funny that Runes and Seraph were both genes and it fit with the Shadowhunter aesthetic, but I'm a huge sucker for biblical allusions/symbolism. The lore hatchlings come with is just kind of a starter/explanation thing and you can do whatever you want with it. I'm actually slowly working through Nephilim (the linked dragon in the first post's) lore to flesh out his own personal story/ some of the origins of the subspecies a bit more.
