It’s the 1st of the month. You’re trying to set up an experiment in which you will need 300x106 cells on the 20th this month.
You know that if you put 600,000 cells into a large tissue culture flask, you will get 10.5x106 cells after 4 days using 25mL of medium containing 10% FBS. This takes 10min per flask. If you use a small tissue culture flask, a seed of 200,000 cells will get you 6.4x106 after 4 days using 15mL of medium containing 10% FBS and taking only 6 minutes.
The longest you can leave cells growing is 4 days - any longer and they will die.
At the moment, you are maintaining this cell line by using a small Tissue Culture flask and harvesting them every 4 days and re-seeding the same flask with 200,000 cells. Today (the 1st) is the day these cells need to be harvested and re-seeded.
