You can't really unequip them, but you can place new abilitystones on top of them. This destroys the old one (you can't resell or reuse it, it's gone), but it means you can replace stuff you don't like. Choose wisely!

well it has to have a glowing box around the space. If it doesn't then that rune or stone won't work for that spot. Certain ones go in certain spots.
