A Nuzlocke Challenge is just a way to make a game more challenging. You give yourself a set of rules that you MUST follow as it's meant to create a higher level of difficulty. Nuzlocke challenges can be used in a wide variety of games, not just with Flight Rising, though I think Pokemon is one of the most popular games for these challenges.

To expand; Nuzlocke because of the Pokemon "Seedot" which evolves into "Nuzleaf." and the creator of the Nuzlocke drew Nuzleaf like "John Locke" from Lost. The name Nuzlocke stuck; as Seedot was the First pokemon ever caught in the start of the very first Nuzlocke. He "died" as a Nuzleaf.
Now, Nuzlocke; as Maltshakes explained. Just means a set of rules you make to make something more challenging for yourself. The first Nuzlocke ever started with pokemon; which the rules were 1. you could only catch the first pokemon you encounter in an area and 2, should the pokemon faint it's considered "dead" and you had to release it.
Nuzleaf died to an explosion attack. It was very dramatic.
