If I was a dragon who had runes I'd probably dedicate my life to working out with Tomo to find out what they mean...

Personally I've always seen Runes as the dragon's divine purpose etched into their skin. By their deity. That's why it glows - it reaches all the way into your soul and bleeds the color of your soul out of your skin.

My Runes dragon knows what his runes means, since he is the one who wrote them himself. In his case, they're a banishment spell.
